A released text box, Gain-of-Function Research Provides Valuable Public Health Data, sets out the case for such work — predicting how pathogens might mutate, informing biosurveillance and countermeasure selection — with one pointed caveat: "The utility of these experiments is most likely reduced, however, in cases where the molecular mechanisms of the disease are not well-understood, such as for coronaviruses."
A second text box gives the US biosafety level definitions, BSL-1 through BSL-4, including the maximum-containment requirements: controlled access, clothing change on entry, shower on exit, full-body air-supplied positive pressure suit, and an isolated zone with dedicated supply, exhaust and decontamination systems.
Figure 1 explains serial passage of bacteria in vitro to increase antibiotic resistance, and notes bacteria may lose other capabilities such as pathogenicity in the process.
Figure 2 explains serial passage of a virus in vivo to increase virulence, using ferrets as the worked example, and notes transmissibility may be lost in exchange.
Figure 3 shows how CRISPR-Cas9 could be used to expand a virus’s host range — using guide RNA to cleave viral DNA coding for surface proteins and supplying a repair template carrying a different sequence, so a virus able to infect some birds becomes able to infect birds and pigs.
